LAS VEGAS — OLN324, a VEGF and Ang-2 inhibitor for wet age-related macular degeneration, achieved improvements in anatomy and retinal fluid that were comparable to faricimab, according to a speaker at Clinical Trials at the Summit.

“In the JADE study, we looked at two doses of OLN324 (Ollin Biosciences), 2 mg and 4 mg, and this was compared to faricimab,” Arshad Khanani, MD, MA, FASRS, of Sierra Eye Associates in Reno, Nevada, told Healio. “The JADE study included patients with neovascular AMD who were treatment-naive as well as DME.”

According to Khanani, who focused on neovascular AMD results of the proof-of-concept trial, patients in the faricimab group had a higher baseline central subfield thickness, but OLN324 and Vabysmo (faricimab, Genentech) resulted in comparable rapid and sustained improvements in anatomy and retinal fluid. However, OLN324 demonstrated “faster and greater” reductions in pigment epithelial detachment thickness vs. faricimab.

“This was also sustained for longer than faricimab,” Khanani told Healio.

All three arms of the study experienced vision gains through week 20, with 82% of those treated with OLN324 reaching 12 weeks of follow-up without re-treatment.

The safety profile for OLN324 was positive, Khanani told Healio, with the JADE study supporting the bispecific antibody as “the only treatment that has demonstrated superior anatomic efficacy over faricimab in a randomized control study.”

Healio previously reported that OLN324 resulted in comparable fluid resolution at 12 weeks vs. faricimab in patients with diabetic macular edema.
